The course B244/07 Genome engineering is being offered by the Graduate School of Health, Aarhus University, spring 2022.
Criteria for participation: University degree in medicine, dentistry, nursing, or Master’s degree in other fields and/or postgraduate research fellows (PhD students and research-year medical students).
Aim: The aim of the course is to:
- Provide students with the state-of-the-art knowledge on CRISPR-Cas9 technology
- Introduce the different types of CRISPR gene editing tools (e.g. base editing, prime editing) and their applications
- Instruct how to design the best CRISPR gRNA with high on-target efficiency and low off-target effect – CRISPR data science
- Instruct how to design CRISPR and donor vectors for gene knockin, tagging
- Introduce CRISPR data science and high-throughput CRISPR computing
- Instruct students how to practically analyze and understand CRISPR editing data with TIDE, ICE and NGS
- Introduce the current progress in CRISPR gene therapy
- Prepare PhD students with essential knowledge and tools for their gene editing projects
Learning outcomes: With the completion of this Genome Engineering course, PhD students are expected to
- Acquire a systematic knowledge on CRISPR gene editing technology, progresses, and applications
- Know how to design the CRISPR gRNA for gene knockout
- Learn how to design the donors for gene knockin
- Independently select the best CRISPR system and strategy for a gene editing project
- Independently analyze and interpret CRISPR gene editing results
- Know the concept, resources and potential of CRISPR data science
- Understand the progresses, but also remaining challenges, in CRISPR gene therapy
Content: This course covers:
- The principle of CRISPR gene editing
- The different genetic manipulation tools derived from CRISPR-Cas9
- Base editing
- Prime editing
- Methods used for quantifying CRISPR editing efficiency
- Ways of delivery CRISPR vectors in vitro and/or in vivo: plasmid, RNP, lentivirus, AAV
- CRISPR data science – on-target activity and off-target effect prediction
- Group teaching on CRISPR vector design for gRNA and donor design for gene knockout, knockin, tagging and single nucleotide mutagenesis
- Group teaching on data analysis
- Introduction to CRISPR gene therapy
Recommended knowledge for participation: Genetics, molecular biology, cell biology, and basic bioinformatics.
